iPSC for Modeling Heart Disease

Cardiovascular diseases are the leading cause of death worldwide, including rheumatic heart disease, coronary heart disease, and congenital heart disease. Much of the current understanding of the molecular mechanisms of human cardiovascular disease comes from genetic animal models and heterologous expression systems. However, the genomes differences and considerable functional species differences in hearts between human and non-human have limited in-depth studies of the molecular basis of many heart diseases. Therefore, there is an urgent need for in vitro cardiac disease models that can accurately reflect human disease phenotypes. iPSC technology is rapidly advancing to make it possible to generate human or even patient-specific disease cell models.

iPSC represents a novel approach that makes it possible to generate human cells that match the patient of interest and that recapitulate the hallmark features of heart disease in a culture dish. hiPSCs can expand into very large numbers and differentiate into various cell types associated with cardiovascular diseases including vascular endothelial cells, cardiomyocytes and smooth muscle cells, allowing researchers to dissect the molecular basis of disease processes within and outside the cardiovascular system.
