Bovine iPSCs

Domesticated cattle are of high economic value for meat and milk production. The availability of bovine PSCs would be useful to promote the application of advanced reproductive techniques in this species. Currently, genetic modification in cattle is achieved through somatic cloning using primarily fibroblasts. But the difficulty and relative inefficiency of gene targeting in somatic cells has limited the development of this approach. Recently, scientists have proposed that it is possible to generate iPSCs from cattle through similar approaches used for mouse or human cells, enabling the use of bovine pluripotent cells for basic or applied purposes. For example, bovine iPSCs with germline capabilities could be used to improve reproductive traits and biotechnology applications.

Since 2011, efforts and progress have been made in establishing bovine iPSCs. Many research groups have used OSKM or OSKM plus NANOG, or added large T antigen factors expressed in lentivirus, retrovirus, or piggyBac vectors to reprogram bovine fetal or adult cells. These cells showed some degree of pluripotency, but their reprogrammed state is incomplete. Overall, the establishment of fully reprogrammed, chimeric-compatible bovine iPSCs with long-term self-renewal capabilities remains a major challenge in the field and deserves further attention.
